Anne was born about 1471 in Grafton Regis, Northamptonshire, England, the daughter of sir Humphrey III Stafford and Catherine Fray.

She died in 1514 in Snape Hall, Bedale, North Yorkshire, England.

Her husband was Richard Neville, who she married in ABT 1490. The place has not been found. Their fifteen known children were John (1493-1543), Margaret (1494-1532), Dorothy (1496-1532), William (1497-c1545), Katherine (1498-?), Elizabeth (1500-?), Susannah (1501-c1560), sir Thomas (1502-1544), Joan (1503-?), Humphrey (1505-?), George (1505-?), Marmaduke (1506-1544), George (1509-c1567), Christopher (1511-?) and Christopher (1513-?).
